Polyunsaturated fatty acids ameliorate aging via redox-telomere-antioncogene axis

Resumen
Los ácidos grasos poliinsaturados (PUFA), un grupo de nutrientes nutritivos y que promueven la salud, mejoran las enfermedades crónicas relacionadas con la edad. Sin embargo, aún no se comprende bien cómo los ácidos grasos poliinsaturados, especialmente los ácidos grasos poliinsaturados Omega-3, ejercen funciones anti-envejecimiento. Aquí vinculamos el aceite de pescado, el ácido docosahexaenoico (DHA) y el ácido araquidónico (AA) a la etiología del envejecimiento a través de un eje redox-telómero-antioncogén basado en ratones envejecidos inducidos por D-galactosa. Tanto el aceite de pescado como los PUFA mejoraron las actividades de la superóxido dismutasa (SOD) y catalasa hepática y las actividades de la SOD cardíaca dentro del rango de 18% -46%, 26% -65% y 19% -58%, respectivamente, mientras que redujeron la actividad cerebral monoamino oxidasa, nivel plasmático de isoprostano F2 y nivel de peroxidación de lípidos cerebrales en un 56% -90%, 20% -79% y 16% -54%, respectivamente. Por tanto, los ácidos grasos poliinsaturados mejoran el proceso de envejecimiento inducido por estrés oxidativo y redox in vivo, que sin embargo no presenta una forma dependiente de la dosis. En particular, tanto los ácidos grasos poliinsaturados como el aceite de pescado inactivaron eficazmente la telomerasa testicular e inhibieron la expresión de la transcriptasa inversa de telomerasa mediada por c-Myc, mientras que losácidos grasos poliinsaturados Omega-3 en lugar de los ácidos grasos poliinsaturados Omega-6 protegieron el hígado y los testículos contra el acortamiento de los telómeros dentro del rango de 13% -25% y 25% -27%, respectivamente. Por lo tanto, los ácidos grasos poliinsaturados Omega-3 pueden inhibir mejor el proceso de envejecimiento inducido por el daño del ADN. Sorprendentemente, solo el DHA suprimió significativamente la vía de senescencia celular evidenciada por la expresión del antioncogén p16 y p53 testicular. Este trabajo proporciona un apoyo evidente para la intercomunicación entre los PUFA, especialmente los ácidos grasos poliinsaturados Omega-3, y el proceso de envejecimiento mediante el mantenimiento de la homeostasis redox in vivo, rescatando la atrición de los telómeros relacionada con la edad y regulando negativamente la expresión del antioncogén.
